چکیده

In this study, we investigate the expression of Brucella abortus proteins in resporise to oxidative stress. The focus is on several proteins thought to be involved in bacterial protection. We show that catalase and Cu-Zn superoxide dismutase are regulated in response to H2O2 or by superoxide, whereas expression of Hsp70 and Hsp60 are not dramatically affected by oxidative stress. The regulation and protective function of periplasmic catalase is studied in more detail. The results demonstrate that periplasmic catalase in B. abortus is regulated at the transcriptional level in response to external hydrogen peroxide, and a catalase deletion mutant is more susceptible than wild type to external H2O2. 1 Primary researcher and author 2 Graduate student who made catalase deletion mutant and did the hydrogen peroxide sensitivity experiments (Fig. 2) ^ Major Professor

Regulation of Brucella abortus catalase.

All aerobic organisms have mechanisms that protect against oxidative compounds. Catalase, peroxidase, superoxide dismutase, glutathione, and thioredoxin are widely distributed in many taxa and constitute elements of a nearly ubiquitous antioxidant metabolic strategy. Interestingly, the regulatory mechanisms that control these elements are rather different depending on the nature of the oxidativ...

The Brucella abortus general stress response system regulates chronic mammalian infection and is controlled by phosphorylation and proteolysis.

BACKGROUND Virulence of pathogenic bacteria is often determined by their ability to adapt to stress. RESULTS The Brucella abortus general stress response (GSR) system is required for chronic mammalian infection and is regulated by phosphorylation and proteolysis. CONCLUSION The B. abortus GSR signaling pathway has multiple layers of post-translational control and is a determinant of chronic...

Brucella abortus HtrA functions as an authentic stress response protease but is not required for wild-type virulence in BALB/c mice.

A second mutation has recently been identified in the previously described Brucella abortus htrA mutant PHE1. As a result of this finding, a new B. abortus htrA mutant, designated RWP11, was constructed to evaluate the biological function of the Brucella HtrA protease. RWP11 is more sensitive to oxidative killing in vitro and less resistant to killing by cultured murine neutrophils and macropha...

Lipid Peroxidation and Antioxidant System in Erythrocytes of Brucella Vaccinated and Challenged Goats

Brucellosis is a reemerging worldwide zoonotic disease and the occurrence of the disease in humans is largely dependent on the animal reservoir [1]. In human it is mainly caused by Brucella melitensis. The highest rate of human infection is mainly in those areas where rates of brucellosis in sheep and goats are high [2,3] as sheep and goats are main reservoir of B. melitensis [4]. The preventio...

Improved Immunogenicity of Tetanus Toxoid by Brucella abortus S19 LPS Adjuvant

Background: Adjuvants are used to increase the immunogenicity of new generation vaccines, especially those based on recombinant proteins. Despite immunostimulatory properties, the use of bacterial lipopolysaccharide (LPS) as an adjuvant has been hampered due to its toxicity and pyrogenicity. Brucella abortus LPS is less toxic and has no pyrogenic properties compared to LPS from other gram nega...
